วิชาห้าพิษ เป็นสุดยอดวิชาของสำนัก บ้านห้าพิษ ซึ่งเจ้าสำนักได้ถ่ายทอดให้กับลูกศิษย์ 5 คน จนมาถึงคนที่ 6 คือ หยางเต๋อ ที่อาจารย์ไม่สามารถถ่ายทอดวิชาให้ได้เนื่องจากมาถึงวาระสุดท้ายของชีวิต สิ่งที่อาจารย์ยังเป็นห่วงอยู่ก็คือ ศิษย์ทั้ง 5 ซึ่งได้รับการถ่ายทอดวิชาไปแล้ว ได้นำไปใช้ในทางที่ผิด ก่อให้เกิดความเดือดร้อนไปทั่วยุทธภพ อาจารย์จึงขอร้องให้ หยางเต๋อไปตามหาศิษย์พี่ทั้ง 5 เพื่อหาทางยับยั้งพวกเขา
